Assay for Ranbp1, a cytosolic protein that binds strongly to Ran complexed with GTP. The top panel shows the specificity of binding; the binding of radioactive Ran-GTP is blocked by excess unlabelled Ran-GTP and unaffected by excess unlabelled Ran-GDP. The bottom panel shows that Ranbp1 is very well conserved evolutionarily. The binding assay detects homologs in extracts from frog (Xenopus), yeast (Schizosaccharomyces pombe) and human (HeLa) cells.
